【摘要】 目的 研究牛膝多糖对小鼠免疫性肝损伤的保护作用。方法 序贯注射卡介苗(BCG)和脂多糖(LPS),诱导小鼠产生免疫性肝损伤模型。在造模过程中,小鼠每日ig 150、300 mg·kg1牛膝多糖,共10 d。比色法测定血清和肝组织中丙氨酸氨基转移酶(ALT )的含量,TBA法和NBT法测定肝匀浆中超氧歧化酶(SOD)的活力以及丙二醛(MDA)的浓度。并计算脾脏及胸腺重量指数。结果 ig 150、300 mg·kg1牛膝多糖后,可明显降低模型小鼠血清和肝组织中升高的ALT的水平,抑制肝匀浆中上升的MDA水平和升高过低的SOD活性,并降低脾脏及胸腺重量指数。结论 牛膝多糖对小鼠免疫性肝损伤有一定的保护作用。

Methods: An immunological liver injury model was induced by administration of lipopolysaaccharide following injection of BCG vaccine into mice for 10 days.ALT levels in serum and liver homogenates ,the activity of SOD and the concentration of MDA in liver homogenates were examined. Results: Ahyranthesbidentata polysaccharides could significantly lower the levels of ALT, MDA and spleen index and increase the activity of SOD. 牛膝(AchyranthesbidentataBL)又名百倍,鸡胶骨,为苋科多年生草本植物,根为药用部分,是我国的一种传统中药。其味苦、酸,性平,入肝肾经。中华人民共和国药典(2005年版)上记载:牛膝有补肝肾、强筋骨、逐瘀通经、引血下行之功效[1]。牛膝根含三萜皂甙、甾体类、糖类、氨基酸、生物碱类和香豆素类化合物等,其中多糖是其重要的活性成分之一,具有调节机体免疫功能、抗生物分子氧化等活性。本课题前期的研究表明,牛膝多糖对四氯化碳所致化学性肝损伤有一定保护作用,本实验进一步观察了牛膝多糖对BCG(卡介苗)+LPS(脂多糖)联合诱导小鼠产生免疫性肝损伤的影响。
