医学论文范文:槐定碱致痫大鼠海马组织超微结构的观察
【摘要】 目的 建立槐定碱(sophoridine,SRI)致痫大鼠癫痫模型,电镜下观察槐定碱致痫大鼠海马组织超微结构的变化,探讨癫痫发病机制。 方法 将30只成年SD大鼠(雌雄不拘)随机分为生理盐水对照组(NS)、槐定碱致痫组(SRI)、苯巴比妥钠﹢槐定碱组(PBS+SRI),每组各10只。腹腔注射大剂量槐定碱(47.83mg/kg)后,观察大鼠行为学变化以及应用电镜技术观察各组大鼠致痫后海马CA1区和CA3区神经元组织病理学和形态学的改变。结果 腹腔注射大剂量槐定碱后,大鼠出现了痫性行为发作;电镜下可见槐定碱致痫大鼠海马CA1区和CA3区神经元核膜双层结构破坏,神经细胞核严重固缩,染色质颗粒状聚集,线粒体肿胀,嵴断裂或消失,毛细血管内皮细胞核固缩。经苯巴比妥钠干预后,槐定碱致痫大鼠海马CA1区和CA3区神经细胞的损伤程度减轻。结论 大剂量槐定碱可导致大鼠在体海马神经细胞损伤,从而诱发癫痫。

Observation on ultrastructure of morphological deformation on hippocampus in SRI induced epileptic rat

[Abstract] Objective To establish a sophoridine (SRI) induced epileptic rats model to observe ultrastructure of morphological deformation on hippocampus in SRI induced epileptic rat.Methods Sprague Dawley rats were randomly divided into saline- control group (n=10), SRI group (n=10) and sodium phenobarbital (PBS)+ SRI group (n=10). Model group was injected with Sophoridien intracerebroventricularly (i.c.v.); saline-control group was injected with physiological saline as model group; sodium Phenobarbital was injected with Phenobarbital after using Sophoridien for 0.5 hour. SRI was intraperitoneally injected in rats and to observe the changes of rat's behavior. Then ultrastructure of morphological deformation of neurons in CA1 and CA3 of hippocampus in saline- control group, SRI group and sodium phenobarbital (PBS) + SRI group rats were observed through electron microscope. Results The kindling model of rats was induced by intraperitoneal injection of SRI. The behavioral seizure was observed, sings of mitochondrial damage, organelle burst and degeneration of synapses were seen at SRI administration; Neurons of PBS+ SRI group were less damages as compared with those of SRI group and most neural structure was undamaged. Conclusion sophoridine can cause hippocampal neurons from being damaged by epileptic seizures in vivo.医.学全.在.线网站www.lindalemus.com
