医学论文范文:间歇性高糖对大鼠肾小管上皮细胞NRK-52EA转分化的影响
【摘要】 目的:探讨间歇性高糖诱导大鼠肾小管上皮细胞株NRK-52EA转分化干预作用。方法:NRK-52E细胞分为空白组、正常葡萄糖组、高糖组、间歇性高糖组,分别作用72 h后,Western blot检测转化生长因子β1(TGF-β1)、Ⅰ型胶原、金属蛋白酶-2(MMP-2)、α平滑肌肌动蛋白(α-SMA)以及甲状旁腺素相关肽(PTHrP)的表达;CM2H2DCFDA试剂盒检测活性氧(ROS)含量。结果:高糖组及间歇性高糖组NRK-52E细胞TGF-β1、Ⅰ型胶原、MMP2、α-SMA以及PTHrP表达均上调;间歇性高糖组TGF-β1、MMP2以及α-SMA表达水平较高糖组高(P均<0.05),其ROS含量也显著高于高糖组(P<0.01)。结论:间歇性高糖能通过氧化应激诱导肾小管导管上皮细胞表达TGF-β1、MMP2,并促进NRK-52EA细胞转分化为α-SMA细胞。

Effects of Fluctuating High Glucose on the Transdifferentiation of Tubule Endothelial Cell Line NRK-52EA in Mouse LIANG Hua-sheng,ZHONG Yu-huaDepartment of Endocrinology,The Ninth Affiliated Hospital,Guangxi Medical University,Beihai,Guangxi 536000,China)Abstract:Objective To investigate the effects of fluctuating high glucose on the transdifferentiation of tubule endothelial cell line NRK-52EA in mouse.Methods NRK-52EA cells were divided into control group,glucose group,high glucose group,fluctuating high glucose group,the expression of transforming growth factorβ1(TGF-β1),typeⅠcollagen,MMP-2,α-SMA and PTHrP were detected with Western blot,and the content of ROS was determined with CM2H2DCFDA kits after treated for 72 h.Results The expression of TGF-β1,typeⅠcollagen,MMP-2,α-SMA and PTHrP were all up-regulation in high glucose group and fluctutating high glucose group;the expression of TGF-β1,MMP-2,α-SMA and the content of ROS in fluctutating high glucose group was significantly higher than that of high glucose group(P<0.05 or P<0.01).Conclusion Fluctuating high glucose could induce the up-regulation of the expression of TGF-β1,MMP-2 in tubule endothelial cells through oxidative stress,and promote the transdifferentiation from NRK-52EAb to α-SMA.

糖尿病血糖间歇波动目前认为对于糖尿病并发症的发生、发展起着重要的作用,其危害性甚至高于持续性血糖升高。肾小管上皮细胞转分化是肾脏纤维化的重要病理基础,既往研究发现间歇性高糖能显著促进内皮细胞、心肌细胞纤维化进程[1-2],高浓度葡萄糖也可诱导肾脏纤维化[3],但间歇性高糖对肾小管导管上皮细胞的研究并不多见。本实验将观察不同条件葡萄糖状态下大鼠肾小管上皮细胞中调控肾小管导管上皮细胞转分化因子的表达,以探讨糖环境对肾小管上皮细胞转分化的影响医.学.全.在.线www.lindalemus.com。
