医学论文范文:辛伐他汀对高糖诱导的肾小管上皮细胞转分化的影响
【摘要】 目的:观察辛伐他汀对高糖诱导的肾小管上皮细胞转分化的作用。方法:大鼠肾小管上皮细胞随机分成三组(n=8):对照组,高糖组和辛伐他汀组。对照组加培养液,高糖和辛伐他汀组在培养液中加葡萄糖,终浓度为30 mmol/L,辛伐他汀组加入终浓度为1 μmol/L辛伐他汀。在6,12,24,48,72 h时用免疫细胞化学和Western blot检测α-SMA的表达。结果:对照组各时间段均无α-SMA表达。高糖组6 h时出现α-SMA阳性表达,48 h达高峰。与高糖组比较,辛伐他汀组各时间点α-SMA的表达均明显减少(P<0.05)。结论:辛伐他汀可抑制高糖诱导的大鼠肾小管上皮细胞转分化。

Effects of Simvastatin on the Transdifferentiation of Renal Tubular Epithelial Cells Induced with High Glucose UANG Xiao-xia,KE Chang-bin,ZHANG Qing-hong,ZHANG Jian-e^ (Department of Nephrology;Department of Anesthesia,Taihe Hospital,Yunyang Medical College,Shiyan,Hubei 442000,China)Abstract:Objective To investigate the effects of simvastatin in renal tubular epithelial cells transdifferentiation induced with high glucose.Methods Cultured renal tubular epithelial cells were randomly divided into three groups(n=8):control group(group C),high glucose group (group D) and simvastatin group(group S),and given culture solution,high glucose(terminal concentration:30 mmoL/L),high glucose(terminal concentration:30 mmoL/L) plus simvastatin(1 μmoL/L), respectively. The expression of α-SMA was detected with immuocytochemistry and Western blot at 6,12,24,48 and 72 hours,respectively.Results The expression of α-SMA was not detected at all the time points.The positive expression of α-SMA in group D was detected at 6 h and reached peak at 48 h.The expression of α-SMA in group S was significantly lower than that of group D at all the time points(P<0.05).Conclusion Simvastatin could inhibit the transdifferentiation of renal tubular epithelial cells induced with high glucose医.学全.在.线网站www.lindalemus.com.

糖尿病肾病肾间质纤维化并进展成终末期肾衰竭是糖尿病危及患者生命的重要原因,其中肌成纤维细胞(Myof)大量增生并分泌平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)是其中心环节[1-2]。辛伐他汀有抗组织纤维化[3-5],改善肾小球硬化、肾脏保护作用,能延缓肾脏病变的进展[6-11]。但是否能够通过抑制肾小管上皮细胞转分化而抑制肾纤维化报道甚少,本文观察辛伐他汀对高糖诱导的肾小管上皮细胞转分化的作用。
1 材料和方法
1.1 主要试剂
α-SMA单克隆抗体(武汉博士德生物工程有限公司),α-SMA免疫细胞化学用二抗、Western blot用HRP标记二抗(北京中杉金桥生物技术有限公司)。
1.2 大鼠肾小管上皮细胞的培养
大鼠肾小管上皮细胞株(NRK-52E)购于ATCC。用含5%胎牛血清的DMEM培养基,在5%CO2、37 ℃的条件下培养。
