医学论文范文:6-OHDA诱导大鼠不同脑区星形胶质细胞反应差异的研究
【摘要】 目的:观察大鼠不同脑区中的星形胶质细胞对6-羟多巴(6-OHDA)局部注射反应的差异。方法:将6-OHDA局部注入大鼠脑内黑质致密部(SNc)、纹状体(CPu)、腹侧被盖区(VTA)和额叶联合皮层(FrA),用免疫组织化学方法观察这些脑区中星形胶质细胞的变化。结果:6-OHDA注入1 d后各脑区星形胶质细胞反应较轻,7 d后反应明显,并持续至14 d;各组相比较以SNc组和VTA组反应明显,而CPu组和FrA组反应较轻。结论:大鼠不同脑区中的星形胶质细胞对6-OHDA局部注射后的反应存在差异。

The reactions of astrocytes in different brain regions in rat model induced by 6-OHDA

[Abstract] Objective:To observe the reactions of astrocytes in different brain regions in rat model induced by 6-OHDA. Methods:The reactive astrocytes in substantia nigra compact part(SNc), caudate putamen(CPu), ventral tegmental area(VTA)and frontal association cortex(FrA) induced by 6-OHDA were detected by immunohistochemical staining. Results:The reactions of astrocytes were hardly detected on day1 after injecting 6-OHDA. From day 7 on wards the reactions were significant. We also found that the reactions of astrocytes in SNc and VTA were more remarkable than that in CPu and FrA. 动物模型常用的神经毒剂,能选择性损伤单胺能神经元[1]。将其注入黑质致密部(SNc)、纹状体(CPu)或前脑内侧束时,会引起黑质DA能神经元的大量退变死亡及纹状体内的DA水平下降。在6-OHDA诱导的PD模型及PD患者脑中都发现CPu和SNc脑区出现明显的胶质细胞反应。将6-OHDA局部注入腹侧被盖区(VTA)或额叶联合皮层(FrA)能否同样诱导其中DA能神经元胞体及末梢的退变死亡,以及能否激活其内的星形胶质细胞?目前未见详细的研究报道。本实验将6-OHDA局部注入大鼠SNc、CPu、VTA和FrA脑区,观察这些脑区中星形胶质细胞反应的情况,借以探讨不同脑区中的星形胶质细胞对6-OHDA反应的差异性,为进一步研究星形胶质细胞参与DA能神经元及其末梢选择性损伤的机制及临床治疗PD等神经退行性疾病提供一定的实验依据。
1 材料与方法
1.1 实验动物和主要试剂 成年雄性SD大鼠48只,体重220 g左右(南通大学实验动物中心提供)。6-OHDA(Sigma公司),小鼠抗GFAP(Chemicon公司),HRP标记的山羊抗小鼠抗体(Chemicon公司),二氨基联苯胺(DAB)(Sigma公司)。
