医学免费论文:加味桃核承气汤对肝纤维化大鼠TGFβ1蛋白表达的影响
【摘要】 [目的]观察加味桃核承气汤对四氯化碳肝纤维化大鼠肝组织转化生长因子β1(TGFβ1)蛋白表达的影响。[方法]用四氯化碳诱导大鼠肝纤维化模型，研究分组为正常对照组、模型对照组、加味桃核承气汤高、中、低剂量治疗组和秋水仙碱对照组;灌胃给药，疗程8周;用免疫组化技术检测肝组织TGFβ1的蛋白表达;检测血清ALT、AST水平;用苦味酸天狼红胶原染色方法进行半定量肝纤维化计分(SSS)。[结果]高、中、低剂量组及秋水仙碱组的TGFβ1的表达水平均低于模型对照组;经加味桃核承气汤治疗后血清ALT、AST水平及肝纤维化SSS计分显著下降。[结论]加味桃核承气汤可抑制肝纤维化大鼠TGFβ1的表达，达到抗肝纤维化作用。

Abstract：[Objective]To study the effects of TGFβ1 of Jiaweitaohechengtang on hepatic fibrosis induced by CCl4 in rats.[Methods]Hepatic fibrosis models were induced by CCl4 in rats.All the experimental rats were divided into six groups： normal control group，model control group，colchicine positive treatment group，highdose，mediumdose，lowdose jiaweitaohechenqitang treatment groups.Drugs were fed on rats for 8 weeks by intragastric administration.Protein expression levels of transfer growth factorsβ1(TGFβ1)were determined with immunohistochemical staining method.Scores of hepatic fibrosis were assessed with semiquantitive scoring system(SSS)through tianlanghong staining.[Results]The protein expression levels of TGFβ1 in the liver tissues in model control group were markedly increased compared with those in normal control group.Scores of hepatic fibrosis and the levels of serum ALT and AST in model control group were higher than those in normal control group，but lower than those in three treatment groups with jiaweitaohechengqitang.There were significant differences between model control group and three jiaweitaohechengqitang treatment groups.[Conclusion]Jiaweitaohechengqitang has therapeutic effects on hepatic fibrosis induced by CCl4 in rats医.学全.在.线网站www.lindalemus.com.

肝纤维化是各种不同原因所致慢性肝病的共同病理特征，任其发展可致肝硬化，抗肝纤维化治疗可阻断其向肝硬化进展。加味桃核承气汤在临床用于肝纤维化治疗中取得了很好的疗效，现对该方作深入研究，为临床推广应用该方提供实验依据。
1 材料与方法
1.1 材料 (1)实验药品：加味桃核承气汤主要由桃仁、桂枝、大黄、甘草、莪术、白术、香附、茯苓等组成，用传统方法浸泡、煎煮、过滤、浓缩，制成煎剂，每毫升含生药2.7g，由浙江中医学院门诊部煎制;秋水仙碱：昆明制药集团股份有限公司。(2)实验动物：SD大鼠，150只，体重160～200g，雌雄各半。由浙江中医药大学动物实验中心提供。(3)主要试剂： 四氯化碳(CCl4)分析纯：上海凌峰化学试剂有限公司橄榄油：上海化学试剂公司;苦味酸、天狼红：美国Sigma公司。兔抗鼠TGFβ1单克隆抗体：武汉博士德公司，二抗：美国Zymed公司;(4)主要仪器：切片机：德国Leca;生物显微镜：日本OLYMPUS CHK;高清晰度彩色病理图文报告分析系统：武汉同济医科大学;电子天平：上海第二天平衡器厂。
