医学免费论文:骨髓基质细胞体外三维培养的研究
【摘要】 目的:建立骨髓基质细胞(BMSC)体外三维培养的方法,并观察其细胞生长形态及生物学特征。方法:制备鼠尾胶原,对BMSC进行三维培养,共聚焦显微镜观察细胞形态,MTT、ALP钙钴、苏丹Ⅲ染色观察BMSC增殖及分化。结果:三维培养的BMSC早期呈现集落生长特点,后逐渐向不同的形态转变,与二维培养相比,细胞增殖峰值提前,且向成骨及脂肪细胞分化能力增强。结论:BMSC在体外三维培养模式下生长良好,能够继续保持其自我更新能力及分化潜能。

Three-dimensional Cultivation of Bone Marrow Stromal Cells in vitroWU Yan,YANG Jian,ZHANG Hong-kao,LIU Ji-jun,LI Dong-sheng (Department of Cardiology;Department of Orthopedics,Renmin Hospital,Yunyang Medical College,Shiyan,Hubei 442000,China)Abstract:Objective To establish a method for three-dimensional cultivation of bone marrow stromal cells(BMSCs) in vitro, and to observe its growth morphology and biological characteristics.Methods BMSC were cultured with prepared collagen in a manner of three-dimensional cultivation.The morphology of BMSCs was observed under confocal microscopy,the proliferation and differentiation of BMSCs were detected with MTT,Ca-co staining and sudan-Ⅲ staining.Results BMSCs grew as colony-like at early stage,and then differentiated to different forms.Compared with two-dimensional cultivation,the peak of proliferation was in advance,and the capacity of differentiation to osteoblast and adipocyte was strengthened.Conclusion Three-dimensional cultivation of BMSCs could maintain their capacity of self-renewal and differentiation potential,and cells could grow very well医.学.全.在.线www.lindalemus.com.

骨髓基质细胞因其在二维培养中具有较强的自我更新能力及多分化潜能,而被认为是基因治疗中合适的种子细胞。但是细胞二维培养与体内环境有较大差别,没有了基质的支持,细胞贴壁生长,可能会失去原有的形态特征及生长分化能力。三维培养系统为细胞提供了立体的生长空间,能更好的模拟细胞在体内的生理状态。本实验在体外模拟体内环境,构建BMSCs三维培养体系,使细胞所处的环境尽可能接近体内,进一步研究BMSCs的生物学性能。
