摘 要:目的 研究心得安外涂所致豚鼠银屑病样动物模型皮损中角蛋白K14,16,17表达水平的变化,从分子水平研究此种动物模型的可行性. 方法 用50g L-1 心得安乳剂外涂豚鼠耳背部皮肤诱导银屑病样动物模型,应用原位杂交法检测银屑病样皮损中角蛋白K14,16,17的表达. 结果 心得安外涂所致豚鼠银屑病样动物模型皮损中角蛋白K14,16表达水平较正常对照组明显增高,K17的水平无明显变化. 结论 心得安外涂所致豚鼠银屑病样动物模型不仅在组织病理学而且在分子水平上与人类银屑病皮损基本一致.

Abstract:AIM To investigate the changes of keratin14,16,17expression in lesions of guinea pig's psoriasis-like ani-mal model caused by topical treatment with propranolol and explore the feasibility of this animal model at molecular level.METHODS Psoriasis-like animal model was established by topical treatment with5%propranolol emulsion on ear's back of guinea pig.Expression of keratin14,16,17in lesions were detected by in situ hybridization.RESULTS Expres-sion of keratin14,16in lesions of treated group were signifi-cantly increased,while expression of K17was not significant-ly different with that in control group.CONCLUSION Guinea pig's psoriasis-like animal model caused by topical treatment with propranolol has high concordance with human psoriatic lesions not only in histopathology,but at molecular level.

银屑病的发病机制极其复杂,其组织病理学的改变主要表现为角质形成细胞过度增殖、炎症细胞聚集和真皮乳头部血管扩张三大特征.其中,角质形成细胞过度增殖常常伴有异常的角化过程,同时出现角蛋白表达的改变.为了能够更好地研究银屑病的发病机制,合适而科学的动物模型显得极为重要.我们从分子水平研究心得安外涂所致豚鼠银屑病样动物模型的可行性.
