Methods: A casecontrol study was conducted. The polymorphism between hCHK2 gene at rs2278022、rs2602431 and rs2970077 position was analyzed by polymerase chain reactionligase detection reaction (PCRLDR) methods in 107 esophageal cancer cases and 204 noncancer controls. The frequencies of genotype and allele and the association between the genetic polymorphisms and cancer risk were analyzed. Results: No significant difference was observed in the frequency of hCHK2 gene at rs2278022、rs2602431 and rs2970077 position genotypes between cases and controls (P>0.05). Conclusion: The polymorphism of hCHK2 gene at rs2278022、rs2602431 and rs2970077 position is unlikely to be associated with the susceptibility of esophageal cancer医.学.全.在.线www.lindalemus.com.
Key words: hCHK2; esophageal cancer; susceptibility; genetic polymorphism随着对细胞周期及其调控的深入认识,学者们提出了“癌症可能是一类细胞周期异常性疾病”的观点[1]。绝大多数肿瘤细胞都存在若干细胞周期调控机制的改变,周期调控机制的异常可导致细胞的持续性增殖,是肿瘤发生的生物学基础。CHK2是近来新发现的一个细胞周期调控蛋白。在发生DNA损伤或复制阻滞后,细胞通过不同途径激活CHK2,进而作用于下游不同的靶蛋白,最终激活G1、S和(或)G2/M期检测点机制,使细胞周期进程发生阻滞,同时激活修复相关基因的转录,促进细胞对损伤进行修复。由于CHK2在细胞周期中具有重要的负调控作用,许多学者认为CHK2起着抑癌基因的作用,它的异常会导致细胞周期检测点对DNA损伤或复制阻滞不能发挥正常功能,从而引起肿瘤易感性增加,最终导致肿瘤的发生或发展。因此,我们采用聚合酶链式反应连接酶检测反应(polymerase chain reactionligase detection reaction,PCRLDR)技术对新疆汉族人群107例食管癌患者和204例非食管癌患者进行了hCHK2 rs2278022、rs2602431和rs2970077基因多态性的检测,并分析探讨了其基因多态性与食管癌遗传易感性之间的关系,以期为食管癌的防治提供理论依据。
1材料与方法
1.1研究对象收集2005~2007年间新发的、由5所医院(新疆医科大学第一附属医院、第二附属医院、附属肿瘤医院、伊犁州新华医院、伊犁州友谊医院)经内窥镜、X线及病理确诊的汉族原发性食管癌患者107例作为病例组,其中男性76例,女性31例,并选择与病例无血缘关系、同民族、同性别、年龄相差±5岁、同期住院的非食管癌患者204例作为对照组,其中男性151例,女性53例。所有研究对象采集外周静脉血4 ml,EDTA抗凝,20℃保存至提取DNA。全部调查对象均知情同意。
